β-环糊精增敏大豆苷元-3′-磺酸钠荧光

摘要 目的 利用β-环糊精对新型药物大豆苷元-3′-磺酸钠的荧光增敏作用建立一种测定其含量的荧光光度分析新方法。方法采用稳态荧光法在λex/λem=310nm/480nm处测定大豆苷元-3′-磺酸钠的相对荧光强度。结果β-环糊精与大豆苷元-3′-磺酸钠在适当条件下产生包络作用,对大豆苷元-3′-磺酸钠的荧光产生明显的增敏作用。所形成包络物的包结比为1:1,包络常数为736。大豆苷元-3′-磺酸钠的荧光强度与其浓度在0~0.40μg/mL的范围内呈良好的线性关系,检测下限为2,26ng/mL。结论基于β-环糊精对大豆苷元-3′-磺酸钠荧光增敏作用而建立的荧光光度分析新方法,在测定其含量时,操作简便、快速、重现性好。 Aim Based on the significant fluorescence enhancement of β-CD on a new drug, sodium daidzein-3′- sulfonate (SDS), to develop a new spectrofluorimetric method for the determination of SDS in the aqueous solution in the presence of β-CD. Methods Determine the relative fluorescence intensity of SDS at λex/λem = 310 nm/480 nm by using a steady-state fluorescence method. Results β-CD formed a complex with SDS on the suitable condition, which lead to the dramatic enhancement of fluorescence intensity of SDS. The inclusion ratio of the β-CD inclusion complex was 1:1 and its inclusion constant was 736. The linear relationship between the fluorescence intensity and SDS concentration was obtained in the range of 0 -0. 40 μg·mL^-1, with a correlation coefficient (r) of 0. 999 7. The detection limit is 2.26 ng·mL^-1. Conclusion The new spectrofluorimetrlc method developed on the significant fluorescence enhancement of β-CD on SDS is simple, rapid and has a good reappearance.
