摘要
目的:观察模拟微重力旋转培养系统(RCCS)对脂肪干细胞(ADSCs)体外培养的影响。方法:将免ADSCs与三维载体PGA相复合后,置于RCCS系统中进行培养,观察ADSCs增殖能力和超微结构的变化。结果:经过RCCs系统的培养,AD- SCs增殖能力明显增强,细胞形态较静置培养组更接近生理情况。结论:采用RCCs系统进行细胞/载体结合物的体外培养,ADSCs增殖能力明显增强,细胞形态更接近生理情况,有助于培养出接近正常软骨生物力学特性的工程化软骨。
Objective:To investigate adipose derived stem cells(ADSCs) proliferation under microgravity rotating culture system. Methods:Expanded and combined with PGA, ADSCs were cultured under the microgravity rotating cell culture system (RCCS). The proliferation of ADSCs was evaluated by MTT assay, and the biological features of the compound were also observed under the scanning electron microscope. Results: The ADSCst proliferation was enhanced markedly, the cellst morphology were near to those under physiology state. Conclusion:Cultured under the RCCS, the proliferation of ADSCs was enhanced markedly, and the amelioration of the ADSCst biological features was also found by the scanning electron microscope.
