二脱氧肌苷对背根神经节神经元突起生长的影响

摘要 目的探讨二脱氧肌苷(didanosine,ddI)对分散培养的胎鼠背根神经节(dorsal root ganglion,DRG)神经元胞体和突起生长的影响。方法分散培养的胎鼠DRG神经元用不同浓度的ddI(1μg/mL,5μg/mL,10μg/mL,20μg/mL)孵育,对分散培养的DRG神经元用微管相关蛋白2(microtubule associated protein2,MAP2)免疫荧光标记后,用共聚焦激光扫描显微镜(confocal laser scanning microscope,CLSM)观察神经元胞体和突起的改变。结果分散培养的胎鼠DRG神经元用ddI孵育3d后,神经元突起的数目减少和长度变短,而神经元的胞体的大小则没有明显变化。结论ddI可直接影响分散培养的胎鼠DRG神经元突起的生长。 Objective To investigate the effects of didanosine (ddI) on the outgrowth of dissociated cultured dorsal root ganglion (DRG) neuronal. Methods Dissociated cultured DRG cells of rat embryos were treated with ddI at different concentrations (1 μg/mL, 5 μg/mL, 10 μg/mL and 20 μg/mL). After 3 days exposure to ddI, the dissociated DRG cells were processed by microtubule associated protein 2 (MAP2) immunofluorescent labeling and the changes of neuronal cell body and neurites were observed by confocal laser scanning microscopy (CLSM). Results The number of neurites in each neuron was decreased and the length of neurites in each neuron was shortened after 3 days treatment with ddI as compared with those in the control group. Alterations of the diameter of neuronal cell bodies were not observed compared with those in the control group. Conclusion ddI directly affects the outgrowth of neurites of the DRG neurons but does not cause neuronal ceil body shrinkage in dissociated DRG ceil cultures.
