摘要
目的探讨砒霜中的有效成分三氧化二砷(ATO)对MRL/lpr自发性狼疮小鼠的疗效及安全性观察。方法20周龄(发病期)MRL/lpr狼疮小鼠24只,设C57正常小鼠24只做参照,随机平分为治疗组和对照组,治疗组ATO 0.4 mg/(kg.d)隔日腹腔注射,对照组隔日腹腔注射同等量生理盐水,疗程为60 d,比较治疗组和对照组在体质量、病死率、血常规上的差异;采用ELISA法检测小鼠血清抗ds-DNA抗体的表达水平。结果MRL/lpr狼疮小鼠中治疗组体质量增长幅度明显高于对照组(P=0.006);累积病死率25%低于对照组(50%);血小板较对照组有所上升(P=0.009);血清抗ds-DNA抗体(OD值)对照组为1.798 5±0.262 1,治疗组为0.918 3±0.067 7,治疗组低于对照组(P<0.01)。C57小鼠未见在对照组和治疗组中有显著性差异。结论ATO可以延长MRL/lpr小鼠动物生存期限,可降低动物的死亡率,降低外周血抗ds-DNA抗体的表达水平,而对C57正常小鼠则无明显作用。
Objective It is to discuss the curative effect and safety observation of Arsenic Trioxide (ATO) on MRL/lpr spontaneous lupus mice. Methods 24 MRL/lpr lupus mice (20 weeks old) and 24 C57 normal mice were randomly equally divided into two groups. Treatment group was treated with ATO 0.4 mg/(kg· d) intraperitoneal injection every other day and control group was treated with isodose normal saline intraperitoneal injection every other day. The course of treatment was 60 d. The difference of body mass, mortality, case fatality and blood routine test between the two groups was compared. The expression level of mouse serum anti - ds- DNA antibody was detected with ELISA method. Results In MRL/lpr lupus mice, the increasing amplitude of body mass in treatment group was obviously higher than that in control group (P = 0. 006 ), the cumulation mortality in treatment group (25 % ) was lower than that in control group (50 % ), the blood platelet count in treatment group was higher than that in control group (P = 0. 009), the OD value of serum anti - ds - DNA antibody in treatment group (0. 918 3 ± 0. 067 7) was lower than that in control group ( 1. 798 5 ± 0. 262 1) (P 〈 0.01). All the indexes in C57 mice had no significant difference between treatment group and control group. Conclusion ATO can prolong the survival time of MRL/lpr mice, lower mortality and lower the expression level of anti - ds - DNA antibody in peripheral blood, but has no obvious effect on C57 normal mice.
