Storage of the Free Living Conchocelis of Porphya Yezoensis Ueda by Encapsulation-Dehydration under Normal and Normal Low Temperature

摘要 用包埋-脱水法在常温和常低温下保存条斑紫菜自由丝状体,探讨了温度、光(暗)和含水量等因素对存活率的影响。结果表明,条斑紫菜自由丝状体在常温或常低温下都可以保存6个月并保持较高的存活率;其中,4℃(暗)保存的最高存活率达到71.5%;而且保存后的丝状体的生长力达到了保存前的水平。包埋-脱水法操作简单,无需贵重设备,在藻类种质保存中有广阔的应用前景。 The free living conchocelis of Porphya yezoensis Ueda were preserved in normal temperature and normal low temperature by using encapsulation - dehydration method. The facters influencing the algal viability, such as different temperature, illumination intensities and the water content of beads were studied. The results showed that the conchocelis were stored for 6 months under normal temperature and normal low temperature and the high viability were obtained. , the algae viability reached the most point at 71.5 % in the temperature of 4℃ (dark) , furthermore, the algae viablity of the conchocelis preserved could reach the normal level. The method of encapsulation - dehydration simplifies the operation procedure and do not need expensive equipments,so it is a potential method for preservation of the algae.
