SLE患者外周血CD4^+CD25^+调节性T细胞细胞因子分泌的改变 被引量:4

Cytokine secretion changes of peripheral CD4^+CD25^+ regulatory T cells in patients with systemic lupus erythematosus

摘要 目的研究活动性系统性红斑狼疮(SLE)患者外周血CD4+CD25+调节性T细胞白细胞介素(IL)-10、转化生长因子(TGF)-β、干扰素(IFN)-γ和IL-4分泌水平的改变。方法免疫磁珠分离活动性SLE患者和正常对照组CD4+CD25+调节性T细胞,流式细胞术(FCM)检测表达IL-10、IL-4或IFN-γ的阳性细胞百分率,体外培养两组CD4+CD25+调节性T细胞,酶联免疫吸附法(ELISA)检测培养细胞上清液中IL-10、TGF-β和IFN-γ的水平。结果在CD4+CD25-和CD4+CD25+T细胞中未发现分泌IL-4的细胞亚群;SLE患者CD4+CD25+T细胞中分泌IL-10的细胞亚群比例较正常人减少(P<0.05);CD4+CD25-和CD4+CD25+T细胞中均存在分泌IFN-γ的细胞亚群,且SLE患者自分泌IFN-γ的细胞亚群比例增高(P<0.05)。SLE患者CD4+CD25+T细胞分泌IL-10和TGF-β较正常人均减少(P<0.05),自分泌IFN-γ增多(P<0.05)。结论活动性SLE患者CD4+CD25+T细胞存在细胞因子分泌功能异常,表现为TGF-β和IL-10分泌降低,IFN-γ产生增多。 Objective To investigate the secretion of interleukin(IL) -10, transforming growth factor(TGF) -β, interferon( IFN )-γ, and IL-4 of peripheral CD^4+CD25^+ regulatory T cells in active systemic lupus erythematosos( SLE ) patients. Methods CD^4+CD25^+ regulatory T cells were separated from peripheral blood samples of SLE patients' with magnetic activated cell sorting (MACS). The positive proprtion of cells expressing IL-10, IL-4 and IFN-γ were measured with flow eytometry(FCM). IL-10, TGF-[5 and IFN-γsecreeted by CD^4+CD25^+ T cells were measured with enzyme-linked immunosorbent assays( ELISA). Results There were no IL-4 secreting subsets in CD^4+CD25^+ and CD^4+CD25^+ T cells. Compared with healthy controls, SLE patients had decreased proportion of lL-10 secreting snbsets in CD^4+CD25^+ T cells( P 〈 0. 05 ). In CD^4+CD25^+ and CD^4+CD25^+ T cells of all subjects, there were IFN-γsecreting subsets, and the proportion of IFN-γ secreting subsets in SLE patients were higher than that in controls( P 〈 0. 05 ). CD^4+CD25^+ T cells in SLE patients secreted higher level of IFN-γ and lower level of IL-10 and TGF-[5 than these in controls( P 〈 0. 05 ). Conclusion CD^4+CD25^+ regulatory T cells in active SLE patients secrete decreased TGF-β, IL-10 and increased IFN-γ.
