摘要
目的通过观察缝隙连接(GJ)阻断剂-甘珀酸(CBX)对戊四氮(PTZ)点燃癫痫大鼠行为和深部脑电活动的影响,探讨GJ在癫痫发展中的作用。方法①健康成年雄性Sprague Dawley(SD)大鼠15只,其中10只每日腹腔注射PTZ[35 mg/(kg.d)]至点燃后,再分别腹腔注射CBX(10mg/kg)(KCBX组)和生理盐水(K组)3 d;作为对照的5只大鼠(NS组)始终每日腹腔注射生理盐水。观察3组大鼠惊厥行为的变化。②另选7只SD大鼠,于海马、杏仁核埋置电极后点燃,行NS、CBX干预,观察深部脑电活动改变。结果①PTZ点燃癫痫大鼠出现自发性抽搐,自发性抽搐次数K组>KCBX组>NS组。②点燃癫痫大鼠脑电表现多种形式的癫痫样波,以阵发性多棘、多棘慢波为特点;CBX干预后,阵发性节律性棘慢波减少,背景节律波幅增高,慢波增多。结论GJ参与癫痫的发生和发展,其阻断剂CBX有潜在的抗惊厥作用。
Objective To explore the effect of carbenoxolone, a gap junction blocker, on seizure and electroencephalogram (EEG) of PTZ-kindled epileptic rats. Methods ① Of the 15 adult male Sprague-Dawley rats used in the study, 10 were intraperitoneally injected with PTZ E35 mg/(kg · d)] to established epilepsy model of kindled rats, and then were intraperitoneally injected with carbenoxolone (CBX, 10 mg/kg) or normal saline (NS). The other 5 rats were in control group. The changes of seizures were analyzed within K group, KCBX group and NS group. ② After putting electrode in the hippocampi and amydaloid nucleus, epilepsy model of kindled rats was established in 7 rats. The changes of brain wave induced by injection of CBX in the PTZ-kindled epileptic rats were analyzed. Results ① Spontaneous seizures appeared in PTZ-kindled epileptic rats. The number of accumulative times of seizures in KCBX group rats was fewer than that in K group rats. ② EEG recordings of the PTZ-kindled epileptic rats displayed various seizure waveforms. The rhythmic spikes or the spike and wave complexes were obvious properties of the PTZ-kindled epileptic rats. After treated with CBX, the rhythmic spike and wave complexes decreased~ meanwhile, the amplitude of background rhythm enlarged and slow waves increased. Conclusion Gap junction and hemichannels are closely involved in epileptogenesis. Gap junction blocker may have an anti-epileptic potential.
