摘要
为了对不同球囊扩张时间的转移基因效率进行比较。方法:用左旋多聚赖氨酸包被导管球囊,然后吸附携带报告基因Lacz的腺病毒载体(Ad5/LacZ),向兔腹主动脉下段定位转移基因。球囊扩张时间分为5分钟、15分钟和30分钟。用定性(X-Gal染色)和定量(β-Gal活性测定)方法检测基因转移效率。结果:5分钟、15分钟和30分钟的基因转移效率无明显差异,十分接近。结论:利用该局部基因转移系统,球囊扩张5分钟即能实现有效的在体。
Objective: The goal of this study was to compare the efficiencies of in vivo gene transfer with different inflation durations of the catheter balloon. Methods: The reporter gene LacZ carried in adenoviral vectors was locally transferred into the lower parts of rabbit abdominal aortic arteries with poly L lysine coated balloon catheter, which was inflated for 5,15 or 30 minutcs, and the efficiencies of gene transfer were qualitatively and quantitatively evaluated with X Gal staining and β Gal activity assay, respectively. Results: The efficiencies of gene transfer with the durations of 5,15 or 30 minutes of balloon inflation showed no significant difference. Conclusion: With the local gene delivery system, local in vivo gene transfer could be effectively achieved with only 5 minutes duration of balloon inflation.
