摘要
目的研究转移生长因子β1(transforminggrowthfactorβ1,TGF-β1)与开角型青光眼的关系。方法用异硫氰酸胍法提取24只新生牛眼小梁网的总RNA,将TGF-β33质粒导入大肠杆菌HB101中充分扩增、BamHI酶切后,片段以α-32-P-dATP标记成cDNA(complementaryDNA)探针,RNA斑点杂交,放射自显影法观察和测定牛眼小梁网的TGF-β1mRNA的表达。结果牛眼小梁网中有TGF-β1mRNA的表达。结论房水中的部分TGF-β1由小梁细胞分泌。此结果可以作为利用分子生物学手段研究TGF-β1mRNA的异常合成、活化、清除及与开角型青光眼病理改变间关系的基础。
Objective To investigate the relationship between transforming growth factor β 1(TGF β 1) and primary open angle glaucoma. Methods Total RNA of 24 new born bovine trabecular tissue was extracted by guanidine isothiocyanate method. The TGF β 33 plasmid was brought into colibacillus HB101 and amplified. After Bam HI enzyme degradation and label with α 32 P dATP, the RNA was hybridized with the complementary DNA (cDNA) probe and examined by autoradiography to determine the expression of messenger RNA (mRNA) for TGF β 1. Results The presence of mRNA for TGF β 1 in bovine trabecular meshwork was confirmed. Conclusion The TGF β 1 present in normal aqueous humor at least partly is derived from the trabecular meshwork. The result offers a basis for understanding the relationship between abnormal synthesis, activation and clearance of TGF β 1 and the pathogenesis of primary open angle glaucoma in molecular biology.
