研究了外源H2O2和IBA处理对黄瓜幼苗外植体不定根发生过程中0~48h时间段内过氧化物醇(POD,peroxidase,EC1.11.1.7)、抗坏血酸过氧化物酶(APX,apoplastic peroxidase,EC1.11.1.11)、过氧化氢酶(CAT,catalas,EC1.11.1.6)和超氧化物歧化酶(SOD,superoxide dismutase,EC1.15.1.1)4种抗氧化酶活性的影响.结果表明,在黄瓜不定根形成的诱导与起始阶段,POD和APX活性变化完全一致,呈逐渐升高的趋势,CAT活性呈现降-升-降-升的变化,SOD活性变化呈对数曲线.H2O2处理显著降低了不定根诱导阶段和起始阶段POD与APX活性,显著降低了诱导阶段CAT和SOD活性;IBA处理对诱导阶段POD和APX活性影响不显著,却显著升高了起始阶段POD和APX活性,对CAT和SOD活性的影响与H2O2处理基本相同.H2O2对不定根形成的作用机理可能与IBA相同.
The activity of four antioxidases, e. g. peroxidase, apoplastic peroxidase, catalas and superoxide dismutase, was investigated during 0-48 hour's formation of adventitious roots of seedling explants in cucumber. The activity of POD and APX coincidentally increased, CAT showed trends of decrease-increase-decrease-increase and SOD showed a logarithm curve in induction and initiation phase. H2O2-treated reduced the activity of POD and APX in induction and initiation phase, and the activity of CAT and SOD in induction phase. However, IBA-treated had no significant effect on POD and APX activity in induction phase, and markedly increased POD and APX activity in initiation phase. Both H2O2-treated and IBA-treated had the same effects on CAT and SOD activity. The results may imply that the mechanism of H2O2 on promoting the formation of adventitious root is the same as IBA.
