睾丸支持细胞和全反式视黄酸诱导骨髓干细胞向精原细胞分化的研究 被引量:2

摘要 目的探讨睾丸支持细胞(sertoli cells,SCs)和全反式视黄酸(all-trans retinoic acid,ATRA)对骨髓干细胞(bone marrow stem cells,BMSCs)向精原细胞分化的影响。方法常规分离12~16d雄性SD子鼠SCs和2月龄雄性SD大鼠BMSCs,用MTT法检测SCs、BMSCs的存活及增殖情况,采用transwellinsert双室培养观察SCs、ATRA对BMSCs向精原细胞方向分化的影响,并用RT-PCR检测精原细胞特异表达基因stra8mRNA的表达情况,用免疫组化S-P法检测c-Kit的表达情况。结果分离后所获取的SCsFasL免疫组织化学染色结果显示,阳性细胞数平均为(93.2±1.0)%;SCs体外培养结果显示,48h增殖达高峰,并维持在该水平至168h。BMSCs体外培养结果显示,144h时增殖达到高峰,并维持在该水平至168h。ATRA组、共培养组、ATRA+共培养组BMSCs都能表达转分化为雄性生殖细胞的分子标志stra8和c-Kit,而对照组不表达。结论SCs可以通过释放可溶性因子促进间接共培养的BMSCs向精原细胞方向分化,ATRA可以促进培养的BMSCs向精原细胞方向分化。
