摘要
目的探讨芹菜素对大鼠睾丸间质细胞(间质细胞)的作用。方法成年S-D雄性大鼠,取下丘脑、腺垂体进行原代细胞培养;睾丸去除被膜后,经Percoll不连续密度梯度离心法分离纯化间质细胞进行原代细胞培养。采用顺序灌流的方式模拟生殖轴的调控以观察不同浓度的芹菜素(10^(-5)M、10^(-7)M、10^(-9)M)对睾丸间质细胞的作用。结果在体外顺序灌流情况下,芹菜素直接作用于间质细胞,呈剂量依赖性和时间依赖性地抑制其睾酮的分泌;芹菜素分别作用于下丘脑和腺垂体时,也呈时间依赖性地抑制间质细胞分泌睾酮,但无剂量依赖性。结论芹菜素能够直接作用于大鼠间质细胞,抑制其分泌睾酮,也能够作用于生殖轴中的下丘脑和腺垂体通过负反馈调节而间接抑制间质细胞分泌睾酮。
Objective To research the effects of Apigenin on Leydig cells in testes of rats. Methods Hypothalamuses and anterior pituitaries of Adult Sprague - Dawley male rats were harvested and primarily cell cultured ; The testes were decapsulated ; the Leydig cells which were isolated from these testes by Percoll discontinuous density gradient were also given primary cell culture. Sequent perfusion method was deployed in vitro to simulate the regulations of reproductive axle in vivo and the effects of different concentrations of Apigenin ( 10^-5M, 10^-7M, 10^-9M) were observed on Leydig cells in testes. Results In condition of sequent perfusion, the Leydig cells were inhibited directly by Apigenin to secrete Testosterone in dose - dependence and time - dependence ; the secreting of testosterone of Leydig cells was also restrained by Apigenin in time - dependence but without in dose - dependence on Hypothalamus and anterior pituitary Conclusions Apigenin can act on Leydig cells directly so that Testosterone secreting will be suppressed; in addition, it affected Hypothalamus and anterior pituitary in reproductive axle and indirectly influenced the secretions of the Leydig cells in negative feedback.
