摘要
目的:探讨P120连环蛋白(P120catenin,P120ctn)及E-钙粘蛋白(E-cad-herin。E-cad)在宫颈鳞癌组织和宫颈鳞癌细胞系(Caski、Siha)中的表达、分布及其生物学意义。方法:免疫组织化学、免疫细胞化学及RT—PCR检测正常宫颈组织、宫颈鳞癌组织、人脐静脉内皮细胞(HECV)、Caski、Siha细胞系中P120ctn及E—cad的表达与分布。结果:①正常宫颈组织中P120cm及E—cad表达在胞膜,胞浆少见;宫颈鳞癌中P120ctn及E—cad胞膜表达降低,出现胞浆表达;②HECV细胞系中P120ctn大部分表达在胞膜,E—cad胞膜表达量较少;Caski、Siha细胞系中P120ctn胞膜表达量减少、胞浆表达量增加,E—cad胞膜胞浆表达都较少;③HECV及Caski、Siha细胞系中都有P120ctn与E—cadmRNA表达,P120ctnmRNA在HECV、Caski、Siha细胞系中无表达量差异,E—cadmRNA在Caski、Siha细胞系中表达量低于HECV。结论:P120ctn及E—cad的异常表达与宫颈鳞癌发生发展密切相关,异常表达与肿瘤细胞分化程度、淋巴结转移有关,与年龄、临床分期无关。
Objective: To research P120 - catenin(P120ctn) and E - cadherin ( E - cad ) abnormal expression, distribution and biologic significance in samples of squamous cervical carcinoma(SCC) and SCC cell lines( Caski ,Siha ). Methods: Expression and distribution of P120ctn and E - cad were examined in SCC tissue, normal cervical epithelia tissue, HECV, Caski and Siha by immuno- histochemical staining ( SP method) , immuocytochemistry(ICC) and RT - PCR.Results: The results showed that①in the normal cervical epithelia tissue, P120ctn and E - cad were stained in the membrane and low expression in the cytoplasm; In SCC, P120ctn and E - cad found in the cytoplasm, and reduced expression in membranous; ② in the cell lines, P120ctn in HECV were found in the membrane, E - cad expressed little in the membrane ; P120ctn in Caski, Siha was mainly expressed in the cytoplasm, E - cad was found little expression in both of the cytoplasm and membranous; ③mRNA of P120ctn and E -cad were expressed in the cell lines (HECV, Caski and Siha) , P120ctn mRNA was not different between normal and cancer cell lines, E - cad mRNA shows low expression in Caski and Siha compared with HECV. Conclusions: Abnormal expression of P120ctn and E - cad were significantly correlated with generation and development of SCC, also correlated with tumor differentiation status and lymph node metastasis, no significant association with clinical stages and age.
