保肝宁对肝纤维化模型大鼠肝功能及血清瘦素水平的影响 被引量:1

摘要 目的:探讨保肝宁抗肝纤维化的作用机制。方法:将50只大鼠随机分为5组,正常对照组、模型组、秋水仙碱组、保肝宁大剂量组、保肝宁中剂量组,各组均10只。采用复合因素建立肝纤维化大鼠模型。采用全自动生化分析仪检测谷丙转氨酶(ALT)、谷草转氨酶(AST),采用同位素放射免疫分析法检测血清瘦素(Leptin)水平。观察保肝宁对肝纤维化模型大鼠ALT、AST、AST/ALT及Leptin水平等各项指标的影响。结果:与正常对照组比较,模型组及各药物组血清ALT、AST、Leptin水平均明显升高,差异均有非常显著性意义(P<0.01),AST/ALT比值模型组升高显著(P<0.01);而保肝宁中剂量组AST/ALT比值较模型组降低(P<0.05);与模型组比较,保肝宁大剂量组、保肝宁中剂量组血清ALT、AST及Leptin水平明显下降,差异有显著性或非常显著性意义(P<0.05,P<0.01),保肝宁大剂量降低AST作用明显优于秋水仙碱,2组比较,差异有显著性意义(P<0.05)。结论:保肝宁可有效防治大鼠肝纤维化,其作用机制可能与通过改善肝功能,降低血清瘦素水平有关。 Objective:To explore the therapeutic mechanism of Bao Can Ning (BGN)for liver cirrhosis.Methods:Fifty rats were equally randomized into 5 groups:normal control group,model control group,colchicine group,and large-and middle-dose BGN groups.Rat model of liver cirrhosis was established.Aalanine transferase(ALT)and aspartate,transferase(AST)levels were examined with auto- matic biochemical apparatus,and the leptin level was detected with isotopic radioim- munoassay.The effect of BGN on ALT,AST,AST/ALT and leptin levels was ob- served.Results:Compared with the normal group,ALT,AST,AST/ALT and leptin levels were increased obviously in the other four groups,the difference being signifi- cant between the two groups(P<0.01).ALT,AST,AST/ALT and leptin levels were obviously decreased in BGN groups as compared with the model group(P<0.01).The effect of large-dose BGN was better on decreasing AST level than that of colchicines(P<0.05).Conclusion:BGN has protective and therapeutic effect for rat liver cirrhosis,and its mechanism may be related with the improvement of hepatic function and the decrease of serum leptin level.
