摘要
试验采用完全随机的2×3×6试验设计,考察在不同pH(5.0、5.5)条件下应用3种底物(SigmaP0109、P8810和P3168),检测不同酶活水平(1000、3000、5000、6000、7000U/g和10000U/g)的植酸酶时,其检测值之间的对比关系。结果表明:不同pH条件下,P0109与P3168的检测结果之间差异均不显著,P8810与P3168的检测结果之间差异均极显著;底物P0109可以完全替代P3168进行植酸酶酶活测定;P8810与P3168的检测结果之间存在线性关系,线性方程为:y(pH5.5,P3168)=0.7649x(pH5.5,P8810)+152.18(R2=0.9963)、y(pH5.0,P3168)=0.8767x(pH5.0,P8810)+34.818(R2=0.9979),T检验分析曲线换算值和验证值结果差异不显著,证明两个线性方程可以作为底物P8810替代P3168进行植酸酶酶活测定的换算公式。
To study the relationship of three substrates (Sigma POLO9, P8810 and P3168)at different pH (5.5,5.0),a completely randomized 2x3x6 design was taken to study the relation of detection values on phytase with different activity ( 1000,3000,5000,6000,7000 U/g and 10000 U/g).The results showed as follows : at different pH (5.5,5.0), there was not significant difference between POLO9 and P3168 ;there was highly significant difference between P8810 and P3168. P3168 could be completely replaced by PO109 in phytase activity test.The test results with substrates P8810 and P3168 respectively had linear relations.The linearity equations were y (pH 5.5, P3168) =0.7649x (pH 5.5, P881 0) + 152.18 (R2=0.9963) and y (pH 5.0, P3168 ) =0.8767x (pH 5.0, P8810 ) +34.818 (R2=0.9979) .The difference of the results between caculation and test was not significant by T test,which proved the linearity equations were feasible.
