摘要
目的:研究中药炎克宁汤剂对输卵管阻塞性不孕模型大鼠输卵管上皮Bcl-2及EGFR mRNA表达的影响。方法:96只8w~12w Wistar雌性大鼠随机分为正常对照组、模型组、妇乐冲剂组、炎克宁低中高剂量组,每组16只。造模后20d开始给药,给药后15d、30d处死各组大鼠。采用原位杂交法检测输卵管上皮细胞Bcl-2及EGFR mRNA的表达。结果:正常对照组输卵管未见有Bcl-2 mRNA表达,妇乐冲剂组与模型组比较15d、30dBcl-2mRNA表达明显升高(P<0.05),EGFR mRNA表达明显下降(P<0.05);炎克宁低剂量组Bcl-2及EGFR mRNA表达与模型组比较无显著差异;炎克宁中剂量组Bcl-2及EGFR mRNA表达与妇乐冲剂组比较无显著差异,与模型组比较有显著性差异(P<0.05);炎克宁高剂量组Bcl-2及EGFR mRNA表达与其他各组比较均有显著差异(P<0.05)。结论:解毒除湿、化瘀止痛治疗输卵管阻塞性不孕模型大鼠,能抑制输卵管细胞凋亡,可能与调节Bcl-2和EGFR mRNA蛋白表达有关。
objective: To observe the effect of Yankening decoction on the expression of Bcl-2 and EGFR mRNA in oviduct epithelium of model rats with tubal obstruction infertility. Methods: 96 rats were randomly divided into 6 groups: model group, normal group, Fule granule control group, Yankening decoction treatment group1 (low dosage), group 2( middle dosage ), group 3(high dosage). The expressions of Bcl-2 and EGFR mRNA in oviduct epithelium were examined by in situ hybridization. Results: Compared with the model group, the expression of Bcl-2 mRNA was increased,the expression of EGFR mRNA was de- creased significantly in Yankening decoction treatment group 1,2,3 and Fule granule control group, and there was significant difference between Yankening decoction treatment group 3 and Fule granule control group. Conclusions: Yankening decoction can reduce apoptosis of oviduct epithelium by regulating the expressions of Bcl-2 and EGFR mRNA.
