HAX-1在SLE患者外周血单一核细胞中的表达及其亚细胞定位 被引量:1

Expression and subcellular localization of HAX-1 in peripheral blood mononuclear cells of patients with systemic lupus erythematosus

摘要 目的检测活动期SLE患者PBMC中抗凋亡基因HAX-1的表达水平,并进行亚细胞定位分析。方法分别采用半定量PCR以及Western印迹试验从mRNA和蛋白水平上检测活动期SLE患者及正常人对照组PBMC中HAX-1的表达,并通过激光共聚焦显微镜观察HAX-1在SLE患者PBMC中的亚细胞定位。结果活动期SLE患者PBMC中HAX-1在mRNA水平以及蛋白水平的表达均明显高于健康正常人(P值分别=0.000、0.007),且其相对表达水平与SLEDAI之间呈线性正相关;HAX-1在SLE患者PBMC中主要分布于线粒体内及内质网。结论HAX-1可能通过抗凋亡机制抑制SLE患者体内淋巴细胞凋亡而导致其过度活化,从而参与SLE疾病发生发展过程。 Objective To detect the expwssion of HSl-associated protein X-1 (HAX-1) and to investigate its subcellular localization in the peripheral blood mononuclear cells (PBMC) of systemic lupus erythematosus (SLE) patients. Methods Semiquantitative reverse transcriptase polymerase chain reaction (RT-PCR) and Western blot were applied to detect the mRNA and protein expressions of HAX-1 respectively in PBMC of active SLE patients, and subcellular localization of HAX-1 in PBMC was observed by laser scanning confocal fluorescence microscopy. Results Compared with the healthy controls, both the mRNA and protein expression levels of HAX-1 in PBMC were significantly increased in active SLE patients (Z = 2.679, P 〈 0.01; Z = 4.556, P = 0.000), and positively correlated with SLE disease activity index (both P 〈 0.01 ). HAX-1 was mainly localized in the mitochondria and endoplasmic reticulum of PBMC. Conclusion HAX-l may take part in the development of SLE by inhibiting the apoptosis of lymphocytes and inducing the over-activation of them in SLE patients.
