蔗渣纤维分解菌的选育及其酶学性质的研究 被引量:2

摘要 从土壤中筛选一株产纤维素酶的绿色木霉(Trichoderma viride)YB—1菌株,经紫外线诱变获得一高活力纤维素酶突变株YB—3,其生长快,产孢子能力强,产生的纤维素酶包括CMC、FPA和β—葡萄糖苷酶的活力分别可达2000,469和1750,(mg·葡萄糖/g·干曲)。研究了碳源、氮源、金属离子和表面活性剂对产酶的影响。最佳培养条件为:起始pH5.5,温度30±1℃,时间60h。正交固体最佳培养基为:复合碳源4.5g(蔗渣:麸皮=6:4),草酸铵0.2g,KH_2PO_40.03g,FeSO_40.006g,加水量1:4。热变性温度范围60℃~70℃,热稳定性较好,有较宽的pH适应范围。是一株具有良好工业应用前景的蔗渣分解菌。 A celluJase producing fungus Trichoderma viride YB-1 was isolated from soil. After treatment with UV, a mutant YB-3 which grew rapidly and produced big colonies was obtained. The CMC, FPA and β-glucosidase activities of culture filtrate of YB-3 were 2000, 469 and 1750 (mg. glucose/g. DM) respectively. Course of the enzyme production and effects of carbon, nitrogen, mentalions and surface active agents on enzyme production were investigated. The optimal culture conditions were as follows, initial pH5. 5, temperature 30±1℃, and cultivation time 60h. The optimum medium for YB-3 were investigated by using orthogonal test, i. e. bagasse 2.7g, wheat bran 1,7g, ammonium oxalate 0.2g, KH2PO4 0.03g, FeSO4, 0.006g, ratio of water 1:4, temperature of thermal denaturation range was 60~70℃; cellulase was stable in wide range pH and had a strong thermal stability. It showed that the hydrolyzing bagasse strain had very good applied prospects in industry.
