高效液相色谱法测定水牛角解毒丸中盐酸小檗碱含量

Determination of Berberine Hydrochloride in Shuiniujiao Detoxicating Pill by HPLC

目的探讨水牛角解毒丸的盐酸小檗碱含量测定方法。方法采用高效液相色谱(HPLC)法,色谱柱为C18柱(250m m×4.6m m,5μm),流动相为乙腈-0.033m ol/L磷酸二氢钾溶液(28∶72),检测波长为345nm,流速为1.0m L/m in,柱温为30℃。结果盐酸小檗碱进样量在0.155～1.553μg(n=7)范围内与峰面积线性关系良好(r=0.9999),平均回收率为105.0%,RSD=2.2%(n=9)。结论HPLC法专属性强、灵敏度高、重现性好、操作简便,可用于水牛角解毒丸的质量控制。

Objective To establish a HPLC method for the determination of berberine hydrochloride in Shuiniujiao Detoxicating Pill. Methods The HPLC analysis was performed on a Diamonsil C18（250 mm×4.6 mm, 5 μm）, with acetonitrile-0.033 mol/L potassium dihydrogen phosphate（28 ： 72）as the mobile phase, the detective wavelength was at 345 nm, the flow rate was 1.0 mL/min. Results Berberine hydrochloride at the concentrations of 0. 155 ～1. 553μg showed the good linear（r=0. 999 9）, the average recovery rate was 105.0%, RSD was 2.2% （n =9）. Conclusion The method is simple, sensitive and reliable. It can be used for the quality control of Shuiniujiao Detoxicating Pill.
