摘要
目的观察丹参酮ⅡA纳米球对基质金属蛋白酶表达的影响。方法将35只新西兰雄性大白兔随机分为3组,动脉损伤模型组(A组),空白纳米粒组(B组)以及丹参酮ⅡA纳米球组(C组),A组与C组每组为15只,B组为5只,然后局部灌注空白纳米粒以及PLGA包载的丹参酮ⅡA纳米球于球囊损伤后的兔颈动脉内,观察内膜增殖以及MMP-2表达的情况。结果局部灌注28d后C组与另两组相比,内膜面积明显减少(P<0.01),MMP-2表达进行IOD值分析,发现C组和A组在同一时期比较,其表达明显减少(P<0.01)。结论丹参酮A纳米粒不仅表现出显著抑制损伤后血管内膜增殖的效应,而且能够明显减少MMP-2的表达。
Objective To test the effect of Tanshinone Ⅱ A nanoparticles on the expression of MMP-2. Methods Thirty five male New Zealand white rabbits were randomly divided into three groups. In group A (15 rabbits), the carotid arteries of the rabbits were stripped. In group B (5 rabbits) and C (15 rabiits), nanoparticles and Tanshinone Ⅱ A nanoparticles were perfused respectively in the denudated arteries of the rabbits. The neointimal areas and the IOD values of MMP-2 secretion were measured. Results A significant reduction of neointimal hyperplasia in group C was found compared to the other two groups in terms of the intimal area and the intima-media ratio. Group C had significant lower IOD values than group A. Conclusion Local administration of nanoparticles with incorporated Tanshinone Ⅱ A not only inhibits neointimal hyperplasia but also inhibits the expression of MMP-2 in stripped arteries.
