PMA诱导小鼠Lewis肺癌细胞节律基因的表达

摘要 目的采用体外节律诱导剂以研究小鼠Lewis肺癌细胞近日节律基因的表达情况。方法采用12-十四酸佛波酯-13-乙酸盐(PMA)刺激体外培养的小鼠Lewis肺癌细胞(LLC),RT-PCR检测不同时间点mPer1的mRNA表达水平。Real-time PCR和RT-PCR检测mClock的mRNA表达水平。结果发现LLC细胞的节律基因mClock、mPer1存在近日节律振荡。结论经PMA诱导后,LLC细胞的节律基因存在近日节律振荡,为体外研究授时因子对近日节律的导引机制提供证据。 Objective To investigate the expression of circadian gene in Lewis lung cancer cells (LLC)in vitro. Methods The cultured LLC cells were stimulated by PMA. The expression of mClock, mPer1 at the indicated times was examined. Results The expression of mClock, mPer1 induced by PMA demonstrated circadian oscillation in LLC. Conclusion PMA can induce the circadian oscillation of clock genes, including mPer1 and mClock, in LLC cells.
