摘要
目的通过检测星形细胞瘤组织中LGI1蛋白的表达水平,探讨其与星形细胞瘤发生、发展、恶性程度及预后的关系。方法应用免疫组织化学技术对50例星形细胞瘤患者肿瘤组织和12例来自于脑外伤内减压手术、高血压脑出血及脑血管畸形手术中所取的正常脑组织中LGI1蛋白的表达进行检测。结果肿瘤组与正常组LGI1蛋白阳性细胞百分率[(28.3±27.11%vs(57.3±22.9)%1比较有显著性差异(P〈0.05);低级别星形细胞瘤和高级别星形细胞瘤中LGI1蛋白阳性细胞百分率[(39.5±28.0)%vs(141±18.2)%]比较有显著性差异(P〈0.05)。50例星形细胞瘤患者中肿瘤复发组与未复发组的LGI1蛋白表达阳性比率(8/27vs18/23)比较有显著性差异(P〈0.05):死亡组与未死亡组的LGI1蛋白表达阳性比率(4/21vs22/29)比较有显著性差异(P〈0,05)。50例星形细胞瘤患者中LGI1蛋白表达阳性组与表达阴性组GOS评分(1.846±1.461vs3.917±1.640)比较有显著性差异(P〈0.05)。结论星形细胞瘤患者肿瘤组织中存在LGI1蛋白分子低表达,推测LGI1蛋白缺失率有可能成为预测脑胶质瘤恶性程度及预后的指标之一。
Objective The expression level of the LGI1 protein in astrocytoma tissues was detected in this study using the streptavidin-peroxidase (SP) immunohistochemical method to further explore the relationship of the expression level with the occurrence and development of astrocytoma and investigate the value of LGI1 protein in evaluating malignancy and predicting prognosis. Methods Tumor tissue samples from 50 astrocytoma cases (tumor group) and samples from normal brain tissues of 12 patients (normal group) who had undergone intracranial decompression for cerebral trauma or operations for hypertensive intracerebral hemorrhage and cerebral vascular malformation were hydrated through graded alcohols, and then the antigen was repaired by microwave and the expression of the LGI 1 protein in above tissues was determined by the SP immunohistochemical method. Results There was significant difference in the percentage for positive cells of the LGI1 protein not only between the tumor group and normal group [(28.3±27.1)% vs (57.3±22.9)%, P〈0.05], but also between low-grade (grade Ⅰ to Ⅱ ) astrocytoma and high-grade (grade Ⅲ to Ⅳ) astrocytoma [(39.5±28.0)% vs (141±18.2)%, P〈0.05]. There was also significant difference in the positive expression rate of the LGI 1 protein not only between the tumor relapse group and non-relapse group in the 50 cases with astrocytoma (8/27 vs 18/23, P〈0.05), but also between the death group and survival group (4/21 vs 22/29, P〈0.05). The difference in Glasgow Outcome Scale (GOS) scores was significant between the positive LGI1 protein expression group and negative expression group among the 50 cases with astrocytoma (1.846±1.461 vs 3.917±1.640, P〈0.05). Conclusion The low expression of the LGI1 protein exists in patients with astrocytoma, speculating that the absent rate of the LGI1 protein may become one of the indicators for the prediction of malignancy and prognosis of cerebral gliomas.
