健肌宁对实验性重症肌无力大鼠胸腺差异表达蛋白谱的影响 被引量:2

Effect of Jianji Ning Recipe on the Differential Expressive Proteins in Thymus with Experimental Autoimmune Myasthenia Gravis

摘要 目的:观察中药复方健肌宁对实验性自身免疫性重症肌无力大鼠胸腺差异蛋白表达的调控作用。方法:制备实验性自身免疫性重症肌无力大鼠模型,随机分为正常组、模型组、中药复方健肌宁组;采用双向电泳—串联质谱法分离和鉴定实验性重症肌无力大鼠模型组和正常组间差异表达蛋白,并观察应用中药复方健肌宁连续干预4周后,差异蛋白表达水平的变化。结果:在已鉴定的3个疾病相关蛋白中,健肌宁对于细胞凋亡双向调节因子、醛还原酶1、真核生物翻译起始因子4B表达水平的有调控作用(P<0.05),这些蛋白与细胞凋亡、蛋白质翻译密切相关。结论:实验性自身免疫性重症肌无力大鼠与正常大鼠间有差异表达的蛋白,中药复方健肌宁可以调节差异蛋白的表达水平。 Objective :To analyze the modulation ofJianji Ning Recipe on differential expressive proteins in thymus of rats with experimental autoimmune myasthenia gravis (EAMG). Methods :Establish the EAMG rats'models, then divide them into normal group,model group and Jianji Ning Recipe group. After therapeutic intervention with above drugs for four consecutive weeks, differential expressive protein levels were analyzed by matrix assisted laser desorption/ionization time -of- flight -time -of-flight mass spectrometry( MALD1 - TOF- TOF). Results :All the three differentially expressed proteins whose functions were correlated to cellular apoptosis and protein translation, were well conducted by Jianji Ning Recipe on the expression of proteins' level compared with model ones( P 〈0.05). Conclusion :The results suggested that there were the differential expressed proteins between EAMG rats and normal rats., Jianfi Ning could effectively regulate differential proteins expressive levels.
