One Method of Structure of Myocardial Infarction Model in Mice

摘要 目的:制备心肌梗死小鼠动物模型。方法:清洁级昆明小鼠20%乌拉坦麻醉,经口气管插管,于左侧胸廓第4肋间开胸,结扎左冠状动脉前降支,观察存活情况、心功能、心肌组织病理变化及心肌梗死面积。结果:完成60例动物模型制作,2周后存活43只;术后2周心功能明显下降(P<0.05);病理切片可见心肌纤维断裂溶解,心肌细胞坏死,有炎性细胞浸润;心肌梗死面积为23.6%~31.2%,平均(27.4±1.3)%。结论:该方法操作简单、创伤小、成功率高、结果可靠。 Objective: To found one model of myocardial infarction in mice. Methods : The mice were anaesthetized by 20% urethane, then intubated through oral tracheal and performed left anterior thoracotomy between the region of fourth and fiveth intercostals, finally ligated the left anterior descending coronary artery(LAD). The survivial rate, heart function, pathologic section and the area of infarction were observed. Results :60 animals'LAD were stitched, 43 survived after two weeks. Heart function decreased( P 〈 0.05). The area of infarction were between 23.6% and 31.2%, its average was (27.4 ± 1.3 ) %. Pathologic section suggested myofibers ruptured or lysed, elenauration or necrosis of myocardiac cells, and infiltration of inflammatory. Conclusion : This method is easy to perform and has little damage to animals, and has high success.
