摘要
在经皮冠状动脉腔内成形术(PTCA)后再狭窄的发病机制中,生长因子因刺激中膜平滑肌细胞增生而发挥了重要作用。碱性成纤维细胞生长因子(bFGF)是一种强效的促分裂因子,为证实球囊损伤是否刺激了碱性成纤维细胞生长因子的基因表达,本实验采用Northernblot方法,以bFGF的cDNA探针与兔颈动脉球囊损伤后的颈动脉中膜平滑肌总RNA杂交。结果显示,颈动脉损伤后其平滑肌中的bFGFmRNA水平低于对照组。从而表明bFGF在再狭窄的发病机制中并不是主要的作用因子。
Growth factors are considered to have played important roles in the pathogenesis of restencr sis after Percutaneous Transluminal Coronary Angioplasty (PTCA) as they can stimulate the proliferation of median smooth muscle cells (SMCs). Basic fibroblast growth factor (bFGF) is one of the most potent mitogen. To reveal whether the gene expression of bFGF in arteries were enhanced or not after balloon injury,the total RNA prepared from rabbits carotid arteries with endothelia denuded were hybridized with a cDNA probe for bFGF. The results showed that the mRNA transcription of bFGF decreased after injury compared with that of uninjured. These data suggested that bFGF might not be the ma jor mitogen responsible for restenosis.
