摘要
目的:研究电离辐射能否导致人结肠癌细胞系HT-29中信号转导子与转录活化子6(STAT6)蛋白活化,从而探讨肿瘤出现放射抗拒的可能机制。方法:用流式细胞仪检测HT-29细胞接受2 Gy射线照射后30 min、1h、2 h细胞内STAT6蛋白的活化情况。结果:HT-29细胞接受射线照射后出现STAT6的活化,在照射后1 h达高峰(P<0.01),2 h后稍高于照射前水平(P<0.05)。结论:射线可以引起HT-29细胞中STAT6蛋白的活化,从而导致细胞出现放射后的凋亡抑制,可能部分解释肿瘤细胞的放射抗拒性。
Objective: To investigate whether ionizing radiation can induce the phosphorylation of STAT6 (Signal Transducer and Activator of Transcription 6) ,which may shed a light on the radioresistance of HT29 cell line. Methods: Flow cytometry was used to detect the phosphorylation of STAT6 in HT29 cells at different time after radiation. Results: Radiation could induce the phosphorylation of STAT6 in HT29 cell line, which reached the peak at 1 h after radiation and was remain a higher level than pre-radiation till 2 h after radiation. Conclusion: Radioresistance of HT29 may be explained partially by the phosphorylation of STAT6 after radiation, which has been demonstrated playing an important role in inhibiting apoptosis in HT29.
