高血压性脑出血肝阳化风证大鼠模型的建立 被引量:9

Establishment of model for hypertensive cerebral hemorrhage of Liver Yang causing wind syndrome in rats

摘要 目的建立一种病证结合动物模型——高血压性脑出血肝阳化风证大鼠模型。方法将45只SD大鼠随机分为正常组、高血压肝阳上亢证组、高血压脑出血肝阳化风证组,采取饮用高渗盐水(1.5%)同时灌服附子汤造成高血压肝阳上亢证模型,在肝阳上亢证模型基础上采取脑内注入Ⅶ型胶原酶法诱导大鼠脑出血,造成高血压性脑出血肝阳化风证,采用症状和体征观察、神经功能评估、组织学检查等方法对所建模型进行评价。结果模型大鼠均出现血压明显升高,烦躁、易激惹、眼结膜深红、头晕、口渴、大便干结等症状或体征,肝阳化风组大鼠并出现脑血肿、脑水肿等出血性脑损伤病理改变与偏瘫等神经缺损体征,差异具有统计学意义(P<0.05或P<0.01),与肝阳上亢组比较血压进一步升高,差异具有统计学意义(P<0.05或P<0.01)。结论该模型能够部分模拟人类高血压性脑出血肝阳化风证的病因病机和临床症候,具有病证结合的特点。 Objective To establish a model of hypertensive cerebral hemorrhage of "Liver Yang causing wind syndrome" in rats. Methods Forty-five SD rats were randomly divided into normal control group, Liver Yang hyperactivity syndrome with hypertension group, Liver Yang causing wind syndrome with hypertensive cerebral hemorrhage group. Liver Yang causing wind syndrome model made by Fuzi decoction and high density Nacl (1.5%), based on which, Liver Yang causing wind syndrome with hypertensive cerebral hemorrhage model was induced by that Ⅻ type collagenase was stereotaxical injected into the globas pallidus in rats to build the ICH model. The models would be evaluated in symptoms and signs observation, neurological functions and histological examination. Result All rats showed obviously high blood pressure dysphoria and aggression, pink eye, dizziness, thirsty and dry stool; and hemorrhagic brain injury showed brain hematoma and edema, etc, and neurolologieal deficits like paralysis in Liver Yang causing wind syndrome with hypertensive cerebral hemorrhage group (P〈0.05 or P〈0.01). Compared with Liver Yang hyperactivity syndrome group, the pressure increased further (P〈0.05 or P〈0.01 ). Conclusion The model can partly mimic the pathogenic and clinical symptom characters of human hypertensive cerebral hemorrhage of Liver Yang causing wind syndrome, and has the features combining both diseases and syndromes.
