摘要
多药抗药性是肿瘤化疗失败的主要原因,在多药耐药基因(mdr-1)检测方法中,PCR具有简便、灵敏、特异性强等特点,我室建立了检测mdr-1mRNA的逆转录/多聚酶链反应(RT/PCR)方法,并用KB耐药细胞对其进行了验证,检测灵敏度达8×104个细胞.在实验过程中摸索了RNA提取及反转录PCR过程的技术关键,为临床mdr-1检测提供了一种实用、稳定的手段。
Multidrug resistance of tumor cells is the main cause of chemotherapy failure.This resistance is related to expression of mdr1 gene. There are many methods to detect the expression of multidrug resistance gene. Among these methods, RT/PCR has higher simplicity, sensitivity and specificity. We have established a RT/PCR method of detecting mdr-1 gene expression, verified it using multidrug resistant KB 200 cells and obtained the minimal detection limit of 8×104 cells. Our study may be useful in evaluating the multidrug resistance at the low expression level of mdr-1 in clinical samples.
