蛋白激酶CK_2-β在腺样囊性癌肺转移细胞中的表达及意义

Expression of Protein Kinase CK_2-β in Lung Metastasis Cell of Adenoid Cystic Carcinoma and Its Significance

目的探讨蛋白激酶CK2-β在唾液腺腺样囊性癌肺转移细胞(SACC-LM)中的表达及其与肺转移的关系。方法利用Western印迹对蛋白激酶CK2-β在SACC-LM和SACC-83细胞核及细胞质中的表达进行检测分析;采用RT-PCR方法检测蛋白激酶CK2-β被不同浓度抑制剂(DRB)处理后nm23-H1的mRNA表达。结果与对照组SACC-83细胞相比,SACC-LM细胞中蛋白激酶CK2-β具有较高的表达;在SACC-LM细胞中,其在细胞核中的表达高于在细胞质中的表达。并且随着蛋白激酶CK2-β被抑制程度的提高,nm23-H1的表达增加。结论蛋白激酶CK2-β的表达与唾液腺腺样囊性癌的肺转移呈正相关。

Objective To investigate the expression of protein kinase CK2-β in salivary adenoid cystic carcinoma of lung metastasis （SACC-LM）,and the relationship between it and lung metastasis.Methods Expression of protein kinase CK2-β in nucleus and plasma of SACC-LM and SACC-83 were assessed by Western bloting method; mRNA expression ofnm23-H, treating by different concentration of protein kinase CK2-β inhibitor were measured with RT-PCR.Result Comparing with SACC-83 cell,higher level of the expression of protein kinase CK2-β in SACC-LM cell was oberseved. And in SACC-LM,level of the expression in nucleus was higher than that in cytoplasm. With the promotion of suppression of protein kinase CK2-β ,expression of nm23-Hzincreased.Coaclusioa The expression of protein kinase CK2-β is positive related to lung metastasis of salivary adenoid cystic carcinoma.
