摘要
目的动态观测急性肺损伤(ALI)大鼠HGF、KGF的变化,并探讨其变化的意义。方法尾静脉注射内毒素(LPS,5mg/kg)建立大鼠ALI模型。36只SD大鼠随机分为对照组(18只)、内毒素组(18只),进行各组动物动脉血气分析,血浆和支气管肺泡灌洗液(BALF)中肝细胞生长因子(HGF)、角细胞生长因子(KGF)含量及BALF中白细胞数量的测定,并进行各组织的病理学光镜检查。结果与对照组相比,ALI组各时间点BALF中HGF、KGF显著增加(P<0.05),且上述改变随观察时间的延长呈加剧趋势。ALI组血清中HGF、KGF在第一时间即明显升高,并且维持在较高的水平(P<0.05)。各时间点差距不显著(P>0.05)。结论ALI组HGF、KGF水平高于对照组,随时间的延长呈加剧趋势。而血清中没有明显变化,提示ALI病程中检测血清细胞因子不能完全反应ALI患者肺内炎症情况,测定肺内因子可能比血清中更有价值。
Objective To investigate the changes in HGF, KGF after acute lung injury(ALl) induced by PLS. Methods Rat model of ALl was used in this study.Rat were randomly divided into the normal control group (n=18),ALI group (n=18).The effects of arterial blood gas,contents of keratinocyte growth factor (KGF) and hepatocyte growth factor (HGF) in plasma and bronchiol alveolar lavage fluid (BALF),and neutrophlis counts,and contents of HGF,KGF in BALF were analyzed by using Enzyme-linked immunoadsordent assay(ELISA).The pathological changes were examined with light microscope in various tissues.Results KGF and HGF in plasma and BALF were significantly higher than those of control group (P〈0.01).Conclusions The HGF and KGF are increased in alveolar space in ALl and may mediate early events in lung repair.They may play an important role in pathogenesis of ALI.KGF and HGF in BALF ameliorating impairment of functions of visceral organs in ALl rat.
