维药买朱尼对骨性关节炎Caspase-3的调节作用

Regulation of Viguv drugt with maizhuni recipe for caspase-3 of osteoavthvitis

探讨维药买朱尼对骨性关节炎大鼠模型膝关节软骨的Caspase-3表达的调节作用。将30只雄性SD大鼠造模后,随机分为正常组,OA模型组,维药买朱尼组。给药4天后,免疫组化法(SP)检测关节软骨Caspase-3蛋白表达,RT-PCR法检测Caspase-3RNA的表达强度。结果:两种方法均显示正常组的Caspase-3弱表达,模型组的Caspase-3强表达。维药买朱尼可以下调Caspase-3表达,与模型组相比有非常显著差异(P<0.01)。结论:维药买朱尼能够下调膝骨性关节炎Caspase-3的表达。

Objectives：To investigate the expression of caspase - 3 in the chondrocyte of a OA rats treated by Maizhuni recipe.Methods：30 male SD rats at 4 weeks of age were randomly divided into a normal group（n = 10） ,a OA group（n = 10）which were subjected to anterior cruciate ligament transsection（ACLT）of the righte knee for 4 weeks and a OA treated by Bushentongluo Recipe（n = 10）. The right knee was removed after three months, transmission electron microscopy was used for the uhrostructure and immunohistechemical and RT- PCR for the expression of caspase - 3 and the level of the caspase - 3 mRNA. Results： The chondrocytes of OA right knee has apoptosis phenomena. Caspase - 3 expression and the level of the caspase - 3 mRNA was significantly increased in the medal group（ P 〈 0.01 ）, and significantly down regulated by the Maizhuni Recipe compared with the OA group（ P 〈 0.01 ）. Conclusion： this study suggests that down - regulation of Caspase - 3 by Maizhuni Recipe may be one of the mechanisms for treatment of OA.
