摘要
PTEN是迄今发现的第一个具有双重特异性磷酸酶活性的抑癌基因,与多种肿瘤的发生和发展密切相关。PTEN通过直接或间接的作用,整合复杂的信号网络系统。该基因可作为脂质磷酸酶,调节磷脂酰肌醇-3,4,5三磷酸水平而调控细胞的生长、分化及凋亡等;也能作为蛋白质磷酸酶,影响黏着斑激酶和SH2包含蛋白的磷酸化,调节细胞的运动、黏附及迁移等多项生物功能。
PTEN is a kind of tumor suppressor gene with dual specificity phosphatase and. it has been demonstrated to closely associated with carcinogenesis and progression of multiple tumors. PTEN may act directly or indirectly to integrate complex signal network system. As lipid phosphatase, PTEN can regulate the level of PIP3 to control the cell growth, differentiation and apoptosis. While as a protein phosphatase, it modulates the phosphorylation of FAK and She, hence influences the eell movement,adhesion, migration and other cell biological functions.
