摘要
[目的]观察亚硒酸钠(Na2SeO3)与硫酸镁(MgSO4)单独及联合作用对二氧化硅(SiO2)致肺泡巨噬细胞(AM)一氧化氮(NO)、一氧化氮合酶(NOS)水平变化的影响,寻找二者的最佳剂量组合。[方法]采用大鼠肺灌洗法纯化获得肺泡巨噬细胞(1×109个/L),同时加入SiO2及不同浓度的MgSO4溶液、Na2SeO3溶液、Na2SeO3+MgSO4溶液,37℃,5%CO2条件下培养18h后,检测NO含量与NOS活性。[结果]与SiO2组比较,Na2SeO3与MgSO4单独及联合使用在体外均可以降低巨噬细胞NO水平及NOS活性(P﹤0.05),Na2SeO3与MgSO4的交互作用具有统计学意义,且以1.0μmol/LNa2SeO3与1.0μmol/LMgSO4联合作用效果最好。[结论]Na2SeO3与MgSO4二者联合应用在体外可明显降低SiO2粉尘所致AM的NO水平和NOS的活性。
[Objective] To observe the independent and joint action of selenium (Na2SeO3) and magnesium (MgSO4) on the changes of nitric oxide (NO) and nitric oxide synthase (NOS) in alveolar macrophage (AM) induced by silica dust. and find the optinlal combination of dose. [ Metheds] Alveolar nlacrophages were obtained by rat lung lavage method (1 ×10^9 cells/L), furthermore, which were cultured with silica and different concentrations of MgSO4 solution. Na2SeO3 solution as well as Na2SeO3 and MgSO4 solution for 18 hours under the condition of 37℃ and 5%CO2. Finally the value of NO and activity of NOS were measured. [Results] When compared with SiO2 group, the independent and joint action of Na2SeO3 and MgSO4 used in vitro could significantly reduce the value of NO and the activity of NOS (P 〈 0.05). Significant interaction between Na2SeO3 and MgSO4 were found in this study, while the optimal effect of joint acuon was Na:SeO3 at the dose of 1.0 μmol/L combined with MgSO4 at the dose of 1.0 μmol/L. [Conclusion] Na,SeO3 and MgSO4 combined used in vitro can reduce NO values and NOS activity in AM which induced bv silica.
