妊娠肝内胆汁淤积症胎盘细胞凋亡及其机制的研究

摘要 目的通过观察妊娠肝内胆汁淤积症(ICP)患者胎盘细胞凋亡及fas基因mRNA在胎盘中的表达,探讨ICP围产儿死亡率升高的原因和机制。方法采用原位末端标记(TUNEL)和原位杂交法分别检测20例ICP患者和20例正常产妇胎盘细胞凋亡和fas基因mRNA表达情况。结果ICP患者胎盘细胞凋亡指数明显高于正常妊娠组,而fas基因mRNA的表达在两组中无显著差异。结论ICP患者围生儿死亡率升高可能与胎盘组织细胞凋亡水平升高有关,但其细胞凋亡的发生可能与fas基因表达无关。 Objective To investigate apoptosis of placental cells and expression of fas gene mRNA in the placentas of the patients with intrahepatic cholestasis of pregnancy (ICP), and to further explore the causes and mechanism of increased prenatal infant mortality of ICP. Methods TdT- mediated dUTP nick end labeling (TUNEL) and in situ hybridization were used in 20 placental samples from patients with ICP to detect apoptosis and fas mRNA and 20 normal placental samples served as controls. Results Apoptosis index placental cells in the ICP group was significantly higher than that in the control group, whereas there was no significant difference in fas mRNA expression between the ICP group and the control group. Conclusions There is probably a relationship between prenatal infant mortality of ICP and placental cell apoptosis; however placenta cell apoptosis is not related with fas mRNA expression.
