快速检测单头柑橘木虱体内黄龙病病原 被引量:9

A simple and effective method for quick detection of Huanglongbing agent from Asiatic psyllid (Diaphorina citri)

摘要 本研究通过采用简易的柑橘木虱制样方法,利用PCR检测技术在单头柑橘木虱体内检测到黄龙病病原,并利用XbaⅠ限制性内切酶将从柑橘木虱体内扩增的黄龙病病原16SrDNA酶切成520bp和640bp两个片段,证实了木虱体内黄龙病病原为亚洲种。本研究成功地建立了一套快速检测柑橘木虱体内黄龙病病原的方法,可为检疫工作者和相关研究人员提供重要参考。 Asiatic citrus psyllid (Diaphorina citri Kuwayama) is currently the only insect vector identified to transmit citrus Huanglongbing (HLB) pathogen and is one of the most important pests in citrus production. Through the feeding process, citrus psyllids acquire the HLB agent from infected sources and transmit the pathogen to healthy trees, eventually leading to a HLB epidemic if no control measure is taken. Detection of HLB agent from citrus psyllid is of particular importance for the disease control. In this study, through adopting a new method for sampling of citrus psyllid and carrying out general PCR, the HLB agent could be detected from single citrus psyllid. After using Xba I to cut the PCR amplify product of the HLB agent's 16S rDNA, two pieces of segment which are 520bp and 640bp could be obtain. The result indicates the agent belongs to Candidatus Liberibacter asiaticus. This method has the advantages of being simple, convenient and fast over the previously described procedures in detection of HLB agent from citrus psyllids.
