大鼠肥厚心肌细胞分离方法的探讨 被引量:1

摘要 目的探讨适用于膜片钳实验急性分离肥厚耐钙SD大鼠心室肌细胞的方法。方法利用自制Langendorff灌流装置,采用四步法获得单个耐钙心肌细胞,以适用于膜片钳离子电流的记录。结果稳定获得30%~40%肥厚耐钙心肌细胞。结论按照本方法在心肌细胞分离过程中的介绍,能获得30%~40%肥厚耐钙心肌细胞。 Objective To investigate the method of stably isolating pachyntic Spraque Dawley rat calcium tolerant ventricular myocytes for patch clamp techniques. Methods Calcium tolerant ventricular myocyte were obtained by four steps with Langendorff apparatus made by ourselves. Ion currens were recorded by patchclamp. Results 30 -40 percents pachyntic calcium tolerant ventricular myocytes can be obtained stably. Conclusion If this method is adopted strictly in the process of isolation ,30 -40 percents pachyntic calcium tolerant ventricular myocytes can be obtained.
