摘要
为了制备出供水稻染色体原位杂交用的染色体标本,以水稻根尖为材料,对8羟基喹啉的预处理时间、纤维素酶和果胶酶的酶解时间以及后低渗时间进行了研究.并将传统的去壁低渗火焰干燥法进行了改进.结果表明:2mmol/L8羟基喹啉预处理1h,5%纤维素酶+5%果胶酶在30℃条件下处理5h,后低渗30min,可获得数量较多。
This study was carried out with root tips of rice as material and the optimal treatment times for root tips were determined with 8hydroxyquinoline, hydrolyzation by Cellulase and Pectinase and posthypotonic period. Results revealed that the speciment with suitable chromosomes for in situ hybridization could be obtained following the procedure of l h 2 mmol/L 8hydroxyquinoline, 5 h Cellulase(5%) and Pectinase (5%) at 30℃ and 30 min in water, respectively.
