从GenBank中搜索到1条来自橡胶树的MYC(Myelocytomatosis)家族基因EST序列,以该序列为保守区,通过RACE技术获得了该基因全长cDNA序列,共1817bp。通过BLAST工具搜索GenBank数据库,结果表明,该cDNA序列同其他植物的MYC家族基因高度同源,认为该cDNA序列为橡胶树MYC家族成员。利用GenScan和Clastal X对该序列进行分析,推测其编码区长度为1428bp,编码476个氨基酸。生物信息学分析结果表明,巴西橡胶树的MYC基因编码的氨基酸序列与其他物种的相似性很高,同时推导的蛋白质序列中还存在着1个螺旋-环-螺旋的DNA结合区域和1个核定位信号,表明克隆到的基因是1个MYC家族成员的转录因子。
An EST sequence of MYC family gene from Hevea brasiliensis was obtained by searching the Genbank databases. The rapid amplification of cDNA end (RACE) technique was employed to amplify the full-length cDNA fragment which was 1 817 bp in length. The results of searching the Genbank by BLAST methods showed that this sequence was highly homologous with the MYC genes of other plants, and was considered as the MYC gene in the rubber tree. The gene sequence was analyzed by the GeneScan and ClastalX software, and a 1 428 bp ORF was observed in this sequence. From this ORF reduced were 473 amino acid residues polypeptides. Bioinformatics analysis showed that the deduced HbMYC1 protein had high homology to the MYC family proteins of other plants, and that there existed a Helix-Loop- Helix DNA binding dimerization domain signature and a nuclear localization signal(NLS) domain in the deduced protein sequence. These results showed that the gene cloned from rubber tree was a transcriptional factor of MYC family.
