摘要
[目的]系统探讨外源生长调节物质对甜玉米幼胚愈伤组织及体细胞再生植株的诱导作用,寻找最佳培养基配方。[方法]以甜玉米甜单10号杂交种幼胚为外植体,以复合型培养基为基本培养条件,应用正交试验设计方法对影响玉米幼胚再生的4个生长调节剂的作用进行探讨,通过DPS软件分析对9种培养基进行数据分析,得出适合实验基因型的最佳培养基,初步建立了甜玉米幼胚再生体系。[结果]AgNO3是诱导Ⅱ型愈伤的关键因素,其次是2,4-D和L-pro,而CH影响最弱。最佳培养基为N6(大量)+B5(微量)+0.1g/L肌醇+500mg/LPro+2mg/L2,4-D+10mg/LAgNO3。[结论]AgNO3对诱导愈伤组织有很大的促进作用,但使用浓度不宜太高,否则会增加畸形苗的比率。
[Objective] The aim of the study was to systematically discuss the induction effects of exogenous growth regulator on the callus of immature embryo of sweet corn and the regenerated plant of somatic cell,and find the optimum medium formula.[Method] With the immature embryo of sweet corn hybrid Tiandan 10 as explant and the compound medium as basic culture condition,the effects of 4 growth regulators influencing the regeneration of immature embryo of sweet corn were discussed by using orthogonal design.Through the data analysis on 9 kinds of media by DPS software,the optimum medium suitable for the experimental genotype was obtained and the regeneration system of the immature embryo of sweet corn was preliminarily established.[Result] AgNO3 was the key factor inducing type Ⅱ callus,the next were 2,4-D and L-pro,the effect of CH was weakest.The optimum medium was N6(macroelement) + B5(microelement) + 0.1 g/L inositol + 500 mg/L Pro + 2 mg/L 2,4-D + 10 mg/L AgNO3.[Conclusion] AgNO3 has very great promoting e
ffect on inducing callus,but its using concentration is not suitable to be too high,otherwise it will increase the rate of deformed seedlings.
