摘要
[目的]探讨枸杞外植体生根培养的最佳配方。[方法]以枸杞新品种宁杞3号离体培养的鲜嫩枸杞苗为研究材料,对常规1/2MS生根培养基的铁元素和有机成分用量增加1倍,配制为改良生根培养基,研究其对枸杞外植体生根培养的效果。[结果]改良生根培养基对宁杞3号外植体生根培养效果非常明显。枸杞外植体的平均生根率在改良生根培养基中为91.7%,而在常规生根培养基中则为65%;接种35d之后,改良培养基上生长的枸杞外植体的平均株高比对照增加了38%,虽然平均根长减少30%,但根粗壮、发达。在改良培养基中培养的枸杞外植体平均单株茎秆鲜重和根部鲜重,分别比常规培养基培养的外植体提高18.1%和8.5%。[结论]改良生根培养基的铁元素和有机养分有利于外植体的生根和成长。
[Objective] The purpose of study was to discuss the optimum formula of rooting medium for wolfberry explants.[Method] With the tender wolfberry seedlings cultured in vitro of the new wolfberry variety Ningqi 3 as tested materials,the dosages of iron element and organic components in conventional 1/2 MS rooting medium were increased once to prepare improved rooting medium and its effect on the rooting culture of wolfberry explants was studied.[Result] The improved rooting medium had very obvious effect on rooting culture of Ningqi 3 explants.The average rooting rate of wolfberry explants on the improved medium was 91.7% and that on the conventional rooting medium was 65%.On the 35th day since inoculation,the average heights of wolfberry explants growing on the improved medium were increased 38% than CK.Though the average root lengths were reduced 30% than CK,their roots were stronger and developed.Average fresh stem weight per plant and fresh root weight of wolfberry explants cultured on the improved medium
were increased 18.1% and 8.5% than that of explants cultured on conventional medium resp.[Conclusion] The iron element and organic nutrients were in favor of rooting and growing of wolfberry explants
