康莱特诱导人肝癌细胞HepG2凋亡的实验研究 被引量:2

摘要 目的探讨康莱特注射液(KLT)诱导肝癌细胞HepG2凋亡的作用机制。方法用不同浓度的KLT培养肝癌细胞HepG2,应用流式细胞仪检测HepG2凋亡细胞的发生,应用ELISA法检测相关凋亡蛋白Bcl-2的变化。结果流式细胞术证实KLT可诱导肝癌细胞HepG2凋亡(P〈0.01),且具有时间、剂量依赖关系。KLT处理组HepG2细胞中Bcl-2蛋白的表达分别为(16.91±0.20)units/ml、(10.78±1.41)units/ml和(7.66±0.64)units/ml,对照组为(22.65±2.83)units/ml。结论KLT可诱导肝癌细胞HepG2凋亡;且KLT诱导HepG2细胞凋亡过程中,Bcl-2蛋白表达的下调对调控细胞凋亡有重要作用。 Objective To study the apoptosis induced by Kanglaite in the hepatocellular carcinoma cell line HepG2. Methods The HepG2 was treated with KLT at different doses. Flow cytometry(FCM) was used to detect apoptosis, and the ELISA was used to detect the apoptosis related change of protein Bcl-2. Results Markedly apoptosis was detected by FCM in HepG2 cells after treatment by KLT and it produced an obvious time-and-dose-dependent apoptosis effect in HepG2 cells. The expressions of Bcl-2 in KLT treated HepG2 cells were ( 16.91 ± 0.20 ) units/ml, ( 10.78 ± 1.41 ) units/rnl and (7.66 ± 0.64 ) units/ml respectively, and in negative KLT treated HepG2 cells were ( 22.65 ± 2.83 ) units/ml. Conclusions KLT can induce the hepatocellular carcinoma cell line HepG2 apoptosis, and Bcl-2 down-expression may probably function as an important regulator in the process of apoptosis induced by KLT.
