摘要
[目的]为地方名优蔬菜品种的保护提供参考。[方法]对杭州市场上10个主要青菜品种绘制AFLP指纹图谱,研究AFLP标记技术在青菜品种上的鉴定作用。[结果]结果表明:最终筛选出的5对引物共扩增出多态性位点454个,平均每对引物扩增位点90个,多态性位点占总扩增位点的比例平均为57.5%;5对引物对10个青菜品种的区分率均达100%。其中1对引物在329 bp有1条明显的特异条带,可用于AFLP指纹鉴定青菜品种"油冬儿"。[结论]该研究为分子标记辅助育种提供了参考依据。
[Objective] The research aimed to provide references for protecting local famous vegetable varieties. [Method] AFLP fingerprintings of 10 main greengrocery varieties in Hangzhou market were drawn and the functions of AFLP marker technology in identifying greengrocery varieties were studied. [ Result] 454 polymorphic loci were amplified by 5 pairs of premiers that were screened out finally, the average site number amplified by each pair of premier was 90 and the average proportion of polyrrorphic loci to total amplification loci was 57.5%. And the differentiation rates of 5 pairs of premiers to 10 greengrocery varieties all reached 100%. Among them, one pair of premiers had a specific band at 329 bp and could be applied in AFLP fingerprint to identify greengrocery varlety "Youdonger". [ Conclusion] This research provided reference basis for molecular marker assisted breeding.
