摘要
目的:研究整合素αvβ3在胶质瘤细胞增殖和侵袭性生长中的作用,为以整合素αvβ3为靶点治疗脑胶质瘤提供更充分的实验依据。方法:采用MTT比色法和PCNA免疫组化检测法,观察不同浓度的整合素αvβ3抗体对体外培养的C6胶质瘤细胞增殖的影响;建立Boyden小室细胞侵袭模型,观察不同浓度的整合素αvβ3抗体对体外培养的C6胶质瘤细胞侵袭能力的影响。结果:整合素αvβ3抗体对体外培养的C6胶质瘤细胞的增殖有明显的抑制作用,其作用表现出明显的量效关系(P<0.05);整合素αvβ3抗体显著降低体外培养的C6胶质瘤细胞的侵袭能力,其作用亦表现出明显的量效关系(P<0.05)。结论:整合素αvβ3对胶质瘤的细胞增殖和侵袭性生长具有促进作用,抗整合素αvβ3有望成为人脑恶性胶质瘤治疗的一种新的有效方法。
Objective: To investigate the effect of integrin αvβ3 on cell proliferation and invasive ability of C6 glioma cells in vitro, so as to provide more experimental bases for brain glioma treatment by anti-αvβ3integrin. Methods: The effect of various concentration of integrin αvβ3 monoclonal antibody on C6 glioma cells proliferation in vitro was detected by MTT method and SABC immunohistochemistry technique. The effect of various concentration of integrin αvβ3 monoclonal antibody on C6 glioma cells invasive ability in vitro was measured by Boyden chamber invasive models. Results: Integrin αvβ3 monoclonal antibody decreased the cell proliferation of C6 glioma cells markedly in vitro in a dose depended manner (P〈0. 05). Integrin αvβ3 monoclonal antibody inhibited the invasive ablity of C6 glioma cells obviously in vitro in a dose depended manner (P〈0. 05). Conclusion : Integrin αvβ3 can positively regulate the cell proliferation and invasive ablity of glioma, and anti-αvβ3 integrin is expected to be a novel and potent method fot the treatment of human malignant gliomas.
