小麦高低分子量谷蛋白亚基分离纯化方法的研究 被引量:3

摘要 以普通小麦为试材,提取麦谷蛋白,分别利用两种不同型号的葡聚糖凝胶Sephadex G-50和Sephadex G-100对高低分子量麦谷蛋白进行层析分离纯化,并采用SDS-PAGE的方法检测分离效果,利用紫外分光光度计法测定各层析峰蛋白含量,计算蛋白回收率。结果表明,Sephadex G-50的蛋白回收率较高,且对低分子量谷蛋白亚基的纯化有一定效果,但不能实现高分子量谷蛋白亚基的有效分离纯化。Sephadex G-100能有效实现小麦高低分子量谷蛋白亚基的分离纯化,但与Sephadex G-50相比,蛋白回收率低。∶: Glutenin was extracted from common wheat. Two types of polydextran gel Sephadex G - 50 and Sephadex G - 100 were used for study of separation and purification of High and Low Molecular Weight Glutenin Subunits by chromatography. The effects were checked by the method of SDS -PAGE. Protein concentration of each fraction was measured by the method of UV spectrophotometer, and the reclaim ratio of protein was calculated. The results showed that protein reclaim ratio of Sephadex G -50 was higher, and it could be used for purification of LMW - GS, while not for HMW - GS. Sephadex G - 100 could separate High Molecular Weight Glutenin Subunits from Low Molecular Weight Glutenin Subunits effectively, but compared to Sephadex G -50, its reclaim ratio of protein was lower.
