地红霉素多晶型对其体外抗菌活性的影响 被引量:2

摘要 目的研究地红霉素两种晶型的体外抗菌活性。方法采用高效液相色谱法测定含量,色谱柱为C18柱,流动相为乙腈-甲醇-pH值为7.5的0.05mol/L磷酸盐缓冲液(44∶19∶37),检测波长205nm,进样量10μL;用抗生素微生物检定法测定其体外抗菌活性。结果地红霉素两种晶型含量基本接近,但体外抗菌活性存在显著差异。结论试验结果为工业生产中选择高活性、低能耗的晶型,以及实现离线与在线原位监控提供了可靠的技术依据。 Objective To study the influences of different molecular conformations of dirithromycin on antimicrobial activity in vitro. Methods Content determination was performed by HPLC. ODS C18 column was used and the mobile phase was acetonitrile-methanol-0, 05 mol/L phosphate buffer (44: 19:37).The wavelength was at 205 nm with injecting amount of 10 μL. The antimierobial activity was detected by microbiologial assay. Results The content in two conformations of dirithromycin was found to be almost the same, but in vitro antibacterial activity existed significant difference between the two conformations. Conclusion The experimental results provide the reliable technical basis for selecting the conformations with high activity and low energy consumption in industrial production and realizing in situ monitor on/off-line.
