姬松茸诱导急性淋巴细胞性白血病细胞凋亡的研究 被引量:3

摘要 目的探讨姬松茸对急性淋巴细胞性白血病细胞增殖及凋亡的影响。方法运用MTT法和彗星电泳法检测姬松茸对急性淋巴细胞性白血病细胞的增殖抑制及促凋亡作用,应用Westernblot法检测WT1蛋白的表达情况。结果姬松茸在0.625~5g/L浓度范围内作用72h以及2.5g/L姬松茸在作用24,48,72h后对急性淋巴细胞性白血病细胞有增殖抑制作用,这种作用呈时间-剂量依赖关系。姬松茸在作用浓度为0.625,1.25,2.5,5g/L时,急性淋巴细胞性白血病细胞凋亡率分别为7.9%,28.4%,44.3%,60.6%,与阴性对照组(1%)差异有显著性;2.5g/L姬松茸在作用24,48,72h后,急性淋巴细胞性白血病细胞凋亡率分别为3.91%,18.72%,40.58%,48,72h作用时间组与阴性对照组(1%)差异有显著性。2.5g/L姬松茸作用72h后的细胞,WT1蛋白的表达较对照组显著减少。结论姬松茸具有对急性淋巴细胞性白血病细胞增殖抑制及促凋亡作用,WT1在此过程中可能发挥着重要的作用。 Objective To investigate the effect of Agaricus blazei Murill on the proliferation and apopt.osis of acute lymphocytic leukemia in vitro. Methods After treatment with Agaricus blazei Murill, cell proliferation was determined with MTr, and apoptosis was measured with comet assay. Expression of WT1 protein in ALL cells was detected with Western blotting. Results After treatment with various concentrations (0.625, 1.25, 2. 5 and 5 g/L) for 72 h or 2. 5 g/L Agaricus blazei Murill for different time(24, 48, 72 h), the growth inhibition rate exhibited significant increases (P 〈0. 05) in a dose - and time - dependent manner and the cell apoptosis rate was increased to 7.9%, 28.4%, 44. 3%, 60. 6% and 3.91% , 18.72% ,40. 58%, respectively. WT1 decreased significantly compared with that of control group after treatment with 2. 5 g/L Agaricus blazei Murill for 72 h. Conclusion Agaricus blazei Murill can inhibit prolifelation and induce apoptosis of acute lymphocytic leukemia in vitro; WT1 may play an important role in the process.
