摘要
目的:考察知母中总皂苷的提取工艺。方法:以知母中菝葜皂苷元为定量检测指标,HPLC法测定菝葜皂苷元的含量;通过正交实验考察了3个因素(提取溶剂的浓度,固液体积比和提取时间)对知母中总皂苷提取率的影响。结果:标准曲线线性范围为0.10~1.10g/L(r=0.9998),平均回收率(98.00±3.43)%。采用索氏提取法筛选的知母中总皂苷的最佳提取丁艺为:提取剂为体积分数50%的乙醇,固液体积比为1:10,提取时间为4h。结论:所确定的知母总皂苷的提取工艺合理可行,值得进一步向工业化生产推广。
Aim: To investigate the extract technology for hydrolysate in timosaponin. Methods:The orthogonal test with three levels of three factors:the extracting time, the solid/liquid ratio and concentration of solvent, using hydrolysate as indexes to evaluate the results. Results: The calibration curve was linear over the range of 0. 10 ~ 1. 10 g/L (r = 0. 999 8 ). The average recovery was ( 98.00 ± 3.43 ) % . The optimal extracting conditions for hydrolysate were 50% ethanol, and solid/liquid ratio of 1 : 10 and extracting time of 4 h. Conclusion :The extract technology for hydrolysate in timosaponin is feasible and reasonable.
