摘要
目的研究鹿茸液对慢性心力衰竭(cHF)大鼠心肌细胞凋亡和Ⅰ、Ⅲ型胶原表达的影响,探讨鹿茸液治疗CHF的作用和机制。方法本实验采用SD大鼠冠状动脉结扎法制备心梗后CHF动物模型,随机分为鹿茸液高、中、低剂量组、卡托普利组、模型组和空白组,给予相应的药物干预,4周后观察心肌细胞凋亡,心间质Ⅰ、Ⅲ型胶原的变化情况。结果鹿茸液各剂量组和卡托普利组CHF大鼠心肌细胞凋亡及Ⅰ、Ⅲ型胶原表达与模型组相比较低,二者具有统计学差异(P〈0.05),鹿茸液中剂量组和卡托普利组之间无统计学差异(P〉0.05),均优于鹿茸液高剂量组和低剂量组(P〈0.05),鹿茸液高剂量组优于低剂量组(P〈0.05)。结论鹿茸液能够减少CHF大鼠心肌细胞凋亡,减少Ⅰ、Ⅲ型胶原沉淀,从而抑制左室重构,具有良好的应用前景。
Objective To study the influence of succus velvet antler on myocardial cell apoptosis and the expression of type Ⅰ/Ⅲcollagen, and to discuss the action and mechanism of succus velvet antler in treating cardiac failure. Methods 130 SD model rats of congestive heart failure (CHF) were established by coronary artery ligation, and divided into six groups in random. Observe the changes of myocardial cell apoptosis and type Ⅰ/Ⅲ collagen. Myocardial cell apoptosis index (AI) was detected by TUNEL. The expressive value of type Ⅰ/Ⅲ collagen was detected by in situ hybridization technique (ISH). Results Succus velvet antler could devalue AI of myocardial cell and inhibit the expression of type Ⅰ/Ⅲ collagen, showing obvious statistic differences compared with model group (P〈0.05); there was no statistic difference between moderate succus velvet antler group and Captopril group. The curative effect of moderate succus velvet antler group was better than that of high dosage group; while the high dosage group was better than the low dosage group. Conclusions Succus velvet antler could inhibit the expression of type Ⅰ/Ⅲ collagen and effectively resist apoptosis at the same time. The results also suggested that the mechanism may be related to reverse ventricular remodeling.
