摘要
目的:探讨胞外基质(ECM)在胸腺发育中的作用及与T细胞发育分化的关系。方法:用抗纤粘连蛋白、层粘连蛋白、Ⅳ型胶原抗体,以免疫组织化学ABC法对所用胸腺切片染色。结果:13d服鼠胸腺内有点状、串珠状ECM阳性区。胸腺内ECM表达,从鼠胚13d到19d渐增强,主要分布于被膜、小叶间隔、血管壁及部分实质细胞间;以后渐减弱且集中在血管壁。结论:ECM表达的变化与胸腺细胞进入胸腺并在胸腺内发育分化最后释放入血有关。
Objective: Investigating the expression and the distribution of extracellular matrix (ECM) in thedeveloping thmpus of mice, so as to analyse the role of ECM in the development of thymus. Methods: Sectionsof thymus of mice were stained with three monoclonal antibodies (anti-FN, -LN, -Collagen type Ⅳ).Immunohistochemical ABC method was employed. Results: On 13-day fetal mice, there were some positive dotsand link-dots of ECM in thyrnus. From 13d to 19d fetal mice, the expression of ECM in thytnus increasedgradually and ECM distributes mainly in capsule, septum, perivascular spaces and some intercellular spaces.After birth, the expression of ECM in thymus decreased and focus on the perivascular spaces. Conclusion: Theresult suggests that ECM may play an important role in the migration, development, differentiation andemigration of T cells.
