摘要 噬菌体展示技术是一种有效的药物靶点筛选工具,可用其筛选中药的药物靶点蛋白。其原理是将外源性蛋白或多肽的DNA序列插入到噬菌体外壳蛋白结构基因的适当位置,使外源基因随外壳蛋白的表达而表达。药物靶点是药物在体内的作用结合位点,是药物筛选和新药研究的基础,良好的药物靶点也是发现性质优良药物的基础。利用现代生物技术为基础的高通量药物筛选技术,进行中药活性组分筛选是实现中药现代化的有效途径,并促进我国天然药物的开发和应用,为中药的研究提供一套完整的筛选和验证方案。 The Phage display technology (PDT) is technology that expresses exogenic polypeptide with shell protein of phage by inserting the exogenic DNA into the shell gene. Drug target is what molecule is bound by the drug inside our body, and also the fundamental of novel drug development as well as drug selection. An ideal drug target is the prerequisite of good drug. The PDT is one of the most effective selection means for drug target, and able to be applied to find the potential target of Chinese medicines. Using the high - through - put technology to analyze the active compounds of Chinese medicines is one of the most effective ways to modernize the Chinese medicines, and it can promote the development and application of natural compounds.
